Wolvaardt scored an unbeaten 115 off just 56 balls - the quickest hundred by a South African in T20Is - as the home side posted a mammoth total of 220-2 in their 20 overs.
It was the highest ever total for the Proteas Women in T20Is, with Sune Luus contributing a fine 81 off 51 balls.
Wolvaardt and Luus was involved in a second wicket partnership of 176, which was also the highest ever stand for the Proteas Women in the shortest format.
Faye Tunnicliffe was out early, for a single, as the hosts lost a wicket in the second over after Wolvaardt had won the toss and elected to bat on a cool, breezy afternoon in Cape Town.
Luus fell in the 18th over which brought Dane van Niekerk to the crease for her first international appearance in four years after reversing her retirement a few months ago.
The 32-year-old had no time to settle with just more than two overs remaining in the innings and struck an unbeaten 21 off just eight balls that included four boundaries.
The Irish started the run-chase in disastrous fashion, losing two wickets in the opening over of the innings as Luus continued her good match with the ball.
The visitors were then never in the match with Luus finishing with figures of 4-22 in her four overs to complete a good allround display.
Ireland were eventuaslly bowled out for just 115 in 18 overs as the Proteas completed a comfortable win.
The Proteas Women can wrap up the three-match series with the second encounter set to take place in Paarl on Sunday.
