The Premier League club will pay an initial fee of 46 million pounds, with a further 4 million pounds in performance-related add-ons. Tottenham have also included a 20 per cent sell-on clause on any future profit from the defender's sale.
The 19-year-old has signed a five-year contract that will keep him at Brighton until 2031, with the club holding the option to extend his stay by a further year.
Brighton head coach Fabian Hurzeler said the club had tracked Vuskovic's progress for some time and believes he has the qualities to develop into a top defender.
"Luka has enjoyed a fantastic start to his career - it's one we've followed closely," Hurzeler said.
"Last season he demonstrated he can play at a very high level and we want to help him build on that within our environment.
"There's been a lot of external noise about Luka joining us, but he is still a young guy who will need time to adjust to the demands of playing for Brighton and the Premier League. We are confident that he will take this in his stride."
Vuskovic originally joined Tottenham from Croatian club Hajduk Split in 2023 as a 16-year-old in a deal worth around 12 million pounds. He officially became a Spurs player in July 2025 before spending last season on loan at Hamburg.
The centre-back enjoyed an outstanding campaign in Germany, earning a place in the Bundesliga Team of the Season after winning four Rookie of the Month awards and the overall Rookie of the Season honour. He also claimed the Bundesliga Goal of the Year award with a spectacular scorpion-kick finish.
Before his move to Hamburg, Vuskovic gained valuable experience on loan at Radomiak Radom in Poland and Belgian side Westerlo, where his performances continued to attract attention from across Europe.
The defender made his senior debut for Hajduk Split just two days after his 16th birthday, becoming one of the youngest players ever to appear in Croatia's top flight.
Despite being a central defender, Vuskovic has shown an eye for goal throughout his career, scoring 16 times in 78 senior club appearances. He has also found the net once in six matches for Croatia.
His rapid rise has continued on the international stage, with Vuskovic becoming Croatia's youngest-ever World Cup player after featuring for his country at the 2026 FIFA World Cup, where he started the group-stage match against England.
