They will meet in their decisive promotion/relegation play-off clash on Saturday.
With only one place available in the top flight, both clubs head into the final round of fixtures knowing the stakes could not be higher.
Milford currently occupy pole position in the mini-league and require only a draw to complete a historic promotion to the Premiership. Magesi, however, must claim all three points if they are to preserve their top-flight status and avoid an immediate return to the Motsepe Foundation Championship.
Going into the final encounter, Milford lead the standings with six points from three matches, while Magesi sit second with four points from the same number of games. Cape Town FC remain on four points but have already completed their campaign.
Although Milford enter as favourites due to their advantageous position, Marques feels Magesi's experience of dealing with high-pressure situations could prove decisive.
"Whoever wins tomorrow will be promoted to the top tier. A draw will also suffice for Milford FC, so the team that manages the stress better under these circumstances will be favoured on Saturday," Marques told KickOff.
"At present, both teams are feeling the pressure and the intensity. The owner of Magesi must be speaking directly with the players and reminding them of the importance of staying in the PSL and what is at stake."
Marques also suggested Milford may be facing different concerns ahead of the encounter.
He added: "Milford players may also be thinking about what promotion means and whether changes to the squad could follow. The mental side becomes crucial in matches like this.
"I have experienced similar situations at Black Leopards and understand the tension involved. Even though Magesi have struggled this season, I believe their experience in these moments will help them win."
