The former Protea captain, who made her last appearance in international cricket in September 2021, had retired after not being selected for the T20 World Cup in 2023 but reversed her decision earlier this year.
Van Nierkerk was picked both the T20I and ODI squad that will take on Ireland at home from 5 December, with three-match series scheduled in both formats.
She has been in good form for Western Province in the both the Pro50 and Pro20 domestic competitions this summer.
Leah Jones, van Nierkerk's captain at Western Province, was included in the 50-over squad and could make her international debut.
Proteas Women Convenor of Selectors, Clinton du Preez, said: "Dane’s return is a result of her notable domestic form, where she has shown consistency and skill. We look forward to her contribution across both formats.
"Leah Jones, along with several other domestic performers, has shown growth and maturity in her game. This is a fantastic opportunity for her to step up to the international level."
The Proteas will look to build on their performances at the recent 50-over World Cup, where they reached the final before losing to host nation India.
The ODI squad to face Ireland marks the start of a new cycle for the group, with all roads leading towards the 2029 edition.
The immediate focus will be on the shortest format with the T20 World Cup set to take place in England in less than eight months.
