Chiefs host AS Simba of the Democratic Republic of Congo at Dobsonville Stadium on Sunday.
The tie is finely balanced following a goalless first leg in Lubumbashi last weekend, but Petersen believes Chiefs will once again rise to the occasion at home, just as they did in the previous round against Kabuscorp.
"We know what's at stake, we want to go to the group stages, and our minds are set on that target," said Petersen, who was officially confirmed as the new Amakhosi skipper this week.
"We came back from Angola after losing 1-0 and turned it around at home before winning on penalties. Congo was a good experience, and getting a draw there was a positive result. I don't see a reason we won't go through."
Despite Chiefs' recent struggles in attack, Petersen remains positive about their defensive resilience, which he believes will be key on Sunday.
"We've been solid at the back since the start of the season. It's important we start strong, stamp our authority early, and make sure we don't concede. Once our attackers regain confidence, the goals will come," he added.
The new skipper will be hoping he inspires his side to victory by leading from the back to ensure they make it to the group stages of the tournament.
